million.
Melbourne, Australia: Amplia Therapeutics Limited (ASX: ATX), (“Amplia” or the “Company”), a
company developing new approaches for the treatment for cancer and fibrosis, is pleased to
announce further progress across its small molecule, focal adhesion kinase (FAK) inhibitor
program and the release of its Appendix 4C Cash Flow Report (attached) for the quarter ending
30 June 2026.
Operations Update
In April, the Company announced that mature data from its ACCENT trial had been presented in
an oral presentation at the prestigious annual meeting of the American Association for Cancer
Research, highlighting the strong clinical potential of its best-in-class FAK inhibitor, narmafotinib,
in combination with standard chemotherapy for metastatic pancreatic cancer. The updated
analysis showed a manageable safety profile with no significant additional toxicity, an 8%
complete response rate (5 of 64 patients) compared with just 0.2% for chemotherapy alone, an
overall response rate of 36% (42% including unconfirmed responses), a disease control rate of
70% versus 50% for chemotherapy alone, and improvements of more than two months in both
median overall survival (11.1 months) and progression-free survival (7.7 months). The data also
demonstrated a trend toward improved survival among patients achieving better treatment
responses, with efficacy exceeding chemotherapy alone across all measures despite narmafotinib
being administered for only 12 days of each 28-day cycle. Based on its favourable tolerability
profile, future studies will evaluate daily dosing, which may further enhance clinical outcomes.
In May the Company reported that it had initiated a Phase 2b study of narmafotinib in advanced
pancreatic cancer to evaluate daily dosing of narmafotinib in combination with the
chemotherapies gemcitabine and Abraxane®, following FDA feedback and as the first stage of a
planned registrational study. The trial will enrol 12 newly diagnosed patients across two dose
cohorts at 3-4 Australian sites, assessing safety, tolerability, pharmacokinetics, efficacy,
biomarkers and fibrosis, with enrolment expected to commence early in the fourth quarter of
2026 and with critical safety data available in the second quarter of 2027. This clinical study has
been accelerated through the redeployment of resources from the AMPLICITY trial where further
recruitment was halted earlier in the year. Including the safety and pharmacokinetic data from
this trial is expected to strengthen the Company’s planned registrational submission to the FDA
in Q2 2027.
Amplia and the Australia New Zealand Gynaecological Oncology Group (ANZGOG) disclosed in
May that they had partnered to launch the PRROSE trial. This trial is an investigator-initiated study
led by Dr Gwo Yaw Ho of Monash Health, to evaluate narmafotinib in combination with standard
chemotherapy for patients with high-grade serous ovarian cancer who have responded poorly to
initial chemotherapy treatment. The trial, which will enrol approximately 20 patients across
Australia and New Zealand, aims to assess safety while exploring whether the addition of
narmafotinib can improve response rates and increase the number of patients eligible for
potentially life-extending surgical resection, addressing a significant unmet medical need. The
study will also incorporate extensive biomarker analyses to better understand narmafotinib’s
mechanism of action in this indication. There is a compelling biological rationale for FAK inhibitors
in ovarian cancer and the Company has preclinical data further supporting the clinical potential
in this indication. The trial represents an important opportunity to improve outcomes for up to
20% of patients with limited treatment options, while also broadening the clinical utility of the
Company’s FAK inhibitor program. Lead investigator Dr Gwo Yaw Ho highlighted ANZGOG’s strong
collaborative network and track record of delivering rigorous, potentially practice-changing
clinical research, noting the trial’s commitment to translating promising science into meaningful
evidence that could inform future treatment options for women with ovarian cancer.
Amplia announced Board leadership changes as part of its Board renewal and succession
strategy, with Dr Warwick Tong retiring as Chairman on 30 June 2026 after serving as a Director
since 2018, while remaining on the Board as a Non-Executive Director. Independent Non-
Executive Director Ms Jane Bell AM, who joined the Board in 2021 has been unanimously
appointed Chair effective 30 June 2026. Dr Tong said it had been “a great privilege” to help shape
the Company over the past eight years and that, given the success of recent clinical trials and
growing interest in Amplia’s portfolio, it was an appropriate time to implement Board changes,
expressing confidence that Ms Bell would bring valuable life sciences experience and strategic
insight to the role. Ms Bell thanked Dr Tong for his “exemplary leadership, expertise and dedicated
service,” noting that his contributions to drug discovery, development, partnerships and
corporate strategy had been integral to Amplia’s progress, and said she looked forward to leading
the Board and executing the Company’s long-term strategy for shareholders.
In early July the Company announced the appointment of experienced biopharmaceutical
executive Brett Carter as a Non-Executive Director further strengthening the Board as the
Company advances narmafotinib into later-stage clinical development. Mr Carter brings more
than 25 years of global experience spanning oncology drug development, corporate strategy and
business development, including 8 years with GSK's global corporate transactions team, where
he led major licensing, acquisition, investment and divestment deals across Europe, Asia and the
United States. He also served as CEO of the Cancer Therapeutics CRC, where Amplia’s drug assets
were originally discovered.
During the quarter a new patent application from the Company, covering chemically novel
inhibitors of focal adhesion kinase (FAK), was published. This patent application, if granted, further
strengthens the Company’s intellectual property portfolio and potentially provides protection for
these new compounds and their therapeutic use through to 2046. The application expands
Amplia’s existing FAK franchise, which already includes its lead clinical-stage candidate
narmafotinib and preclinical candidate AMP886, by introducing additional compounds closely
related to narmafotinib that demonstrate potent and selective FAK inhibition. These novel
molecules broaden the Company’s development pipeline, create opportunities for future
development across additional disease indications, and enhance the long-term commercial value
of its assets.
Financial update
Amplia finished the June 2026 quarter with a cash position of $24.0 million (March 2026: $27.9
million). During the quarter, the Company had net operating cash outflows of $3.9 million in
relation to operating activities (March 2026: $3.5 million inflows). Operating cashflows included:
• Outflows of $1.1 million for staff and administration/corporate costs; and
• Outflows of $3.0 million for research and development costs, which primarily related to
trial costs, Contract Research Organisation (CRO), manufacturing and other CMC related
costs incurred in relation to the ACCENT Phase 2 clinical trial.
Payments to Related Entities
In accordance with Listing Rule 4.7C, payments made to related parties and their associates
included in item 6.1 of the Appendix 4C incorporates directors’ fees, salaries and superannuation.
Total payments made for the quarter equals $112,500 and relate to payments to the
CEO/Managing Director in line with the employment contract and payments to the Non-Executive
Directors.

About Narmafotinib
Narmafotinib (AMP945) is the company’s best-in-class inhibitor of FAK, a protein over-expressed in multiple
cancers and a drug target gaining increasing attention for its role in solid tumours. The drug, which is a
highly potent and selective inhibitor of FAK, has shown promising data in a range of preclinical cancer
studies. Narmafotinib is currently being investigated in the ACCENT trial in advanced pancreatic cancer
where it is dosed in combination with the chemotherapies gemcitabine and nab-paclitaxel (Abraxane®
) in
first-line patients. The trial has achieved its desired outcome in achieving a response rate of 36%, superior
to chemotherapy alone as reported for the benchmark MPACT study. A median Overall Survival (mOS) of
11.1 months has been reported along with an unprecedented complete response rate of 7.8%. A second
trial (AMPLICITY) is being run at sites in Australia investigating the combination of narmafotinib with the
chemotherapy mFOLFIRINOX in advanced pancreatic cancer patients, and a third trial in ovarian cancer (the
PRROSE trial) is scheduled to begin recruiting later this year.
