Melbourne, Australia: Amplia Therapeutics Limited (ASX: ATX), (“Amplia” or the “Company”), developing
new drugs for the treatment for cancer and fibrosis, reports progress in the ongoing development of
narmafotinib along with the release of its Appendix 4C Cash Flow Report (attached) for the quarter ending
31 March 2026.
Operations Update
The Company continues to make significant progress in the development of its best-in-class FAK inhibitor,
narmafotinib, for the treatment of metastatic pancreatic cancer.
Clinical Trial Updates
Over the last quarter the most significant update was provided to the market in March this year. The
Company reported mature results from the ACCENT clinical trial which combines narmafotinib with the
chemotherapies gemcitabine and Abraxane®
. The mature data analysis was derived from a blinded
independent review of clinical response data (specifically Computed Tomography (CT) scans) available up
to mid-March 2026 by a specialist, globally recognised, contract laboratory.
This analysis demonstrated that the combination of narmafotinib with chemotherapy shows a median
overall survival of 11.1 months, more than two months improved over chemotherapy alone in the
benchmark MPACT study. In addition, an unprecedented five confirmed complete responses (CRs) were
recorded, giving a CR rate of 7.8% (5/64). The updated Objective Response Rate (ORR) is 35.9% (23/64), with
four patients still enrolled as of March 2026. Notably, narmafotinib is well tolerated by patients and does
not contribute significant additional toxicity beyond that already associated with the chemotherapy
regimen. Amplia was selected to present these data at the prestigious American Association of Cancer
Research annual meeting in April 2026 as an oral presentation in the Advances in Precision Oncology
symposium.
The Company also presented data from the ACCENT trial in January at the American Society of Clinical
Oncology Gastrointestinal Cancers Symposium (ASCO-GI) meeting in San Francisco. This is a specialist
conference where oncologists and drug developers in the GI cancer space discuss latest data and trends in
the treatment of pancreatic and other cancers.
Conference participation and presentations at these and other international meetings is an important
phase of our engagement and interaction with collaborators, key opinion leaders and potential commercial
partners. This is particularly relevant at this time given the significant developments in the treatment of
pancreatic cancer, most notably with the exciting clinical progress of targeted (personalised) therapies. By
detailing the promising safety, tolerability and efficacy data for narmafotinib observed in the ACCENT trial,
we demonstrate the drug’s significant clinical opportunity in pancreatic cancer when combined with
chemotherapy or targeted therapies.
The Company’s second clinical trial, AMPLICITY, is exploring the combination of narmafotinib with the
chemotherapy regimen FOLFIRINOX. In February, it was announced that two US sites, University of
California, Irvine and The Cleveland Clinic, had completed pre-trial activities and would soon begin
recruitment for the AMPLICITY trial. These sites joined two existing Australian sites, Epworth Hospital
(Melbourne) and Genesis Care (Sydney). In April the Company announced that recruitment to the trial would
be discontinued after three cases of dose-limiting toxicity had been observed, in each case related to the
chemotherapy and not related to narmafotinib. Five patients remain on study at the two Australian sites
and further updates from this study will be reported in due course.
Regulatory and Chemistry Manufacturing and Control (CMC) Updates
In January the Company announced completion of large-scale production of narmafotinib meeting required
purity and quality standards – a major milestone toward Phase 3 readiness. The Company successfully
transitioned manufacturing from R&D to a commercial environment in partnership with its Contract
Development and Manufacturing Organization (CDMO), achieving greater efficiency and cost savings.
Approximately 13 kg of GMP-produced active pharmaceutical ingredient will be subsequently formulated
into oral capsules for current and future clinical trials. Manufacturing complies with internationally
recognised Good Manufacturing Practice (GMP) standards, ensuring product safety and quality.
The Company continues to refine the clinical plan for the registrational study for narmafotinib in pancreatic
cancer. Our regulatory team are working closely with our advisers with the aim of a formal meeting
engagement with the US FDA mid-year.
Preclinical Updates
In March, the Company announced that new data on narmafotinib was presented at the AACR Special
Conference in Cancer Research: RAS Oncogenesis and Therapeutics in Los Angeles on March 6. The preclinical
findings show narmafotinib boosts the effectiveness of kRAS inhibitors in cancer models by blocking
resistance pathways. kRAS inhibitors, currently in development for cancers such as lung, colon, and
pancreatic, are being studied worldwide, but side-effects and resistance remain challenges despite
encouraging mid-stage clinical data.
Operational Updates
In February the Company announced the appointment of Mr Hamish George as Chief Financial Officer
effective 25 February 2026. Mr George succeeds Mr Tim Luscombe of Bio101 Financial Advisory. Mr George
is a Chartered Accountant with degrees from the University of Melbourne and RMIT, previously acted as
Amplia’s CFO, and has extensive CFO experience with a range of life sciences and ASX-listed companies.
Future Outlook
The ACCENT and AMPLICITY trials are ongoing and further updates will be reported as significant findings
become available. Planning for new clinical studies with narmafotinib is well underway, including the
registration-enabling study of narmafotinib in combination with gemcitabine and Abraxane®
.Business
Development activities are also progressing in light of the promising data from the ACCENT study.
Financial update
Amplia finished the March 2026 quarter with a cash position of $27.9 million (December 2025: $31.5
million). During the quarter, the Company had net operating cash outflows of $3.5 million in relation to
operating activities (December 2025: $2.4 million inflows). Operating cashflows included:
• Outflows of $0.9 million for staff and administration/corporate costs; and
• Outflows of $2.8 million for research and development costs, which primarily related to trial costs,
Contract Research Organisation (CRO), manufacturing and other CMC related costs incurred in
relation to the ACCENT and AMPLICITY clinical trials.
Payments to Related Entities
In accordance with Listing Rule 4.7C, payments made to related parties and their associates included in
item 6.1 of the Appendix 4C incorporates directors’ fees, salaries and superannuation. Total payments
made for the quarter equals $147,500 and relate to payments to the CEO/Managing Director in line with

About Narmafotinib
Narmafotinib (AMP945) is the company’s best-in-class inhibitor of the protein FAK, a protein over-
expressed in pancreatic cancer and a drug target gaining increasing attention for its role in solid
tumours. The drug, which is a highly potent and selective inhibitor of FAK, has shown promising data in
a range of preclinical cancer studies. Narmafotinib is currently undergoing a clinical trial (the ACCENT
trial) where it is dosed in combination with the chemotherapies gemcitabine and Abraxane in first-
line patients with advanced pancreatic cancer. The trial has already achieved its desired outcome in
achieving a response rate of 31%, superior to chemotherapy alone and an interim PFS of 7.7 months
has been reported. A second trial – AMPLICITY – has recently opened and is being run under an IND at
two sites in Australia, investigating the combination of narmafotinib with the chemotherapy FOLFIRINOX
in advanced pancreatic cancer patients.
